How long does Vardenafil Hydrochloride stay in your body?
The elimination half-life of vardenafil hydrochloride is about 4 to 5 hours — the time it takes the body to clear about half of it. As a rule of thumb, a medicine is mostly gone after roughly 4 to 5 half-lives, though this varies from person to person with age and kidney or liver function. The label explicitly calls this the TERMINAL half-life (elimination phase), so it is the right number, not a distribution phase. Active metabolite: vardenafil's major circulating metabolite M1 has the SAME ~4-5 h terminal half-life (it does not outlast the parent) and contributes only ~7% of total pharmacologic activity, so the parent number is representative. Populations: exposure (AUC) rises even though the label does not restate a longer half-life for age or organ impairment — AUC is 52% higher in men >=65, 160% higher in moderate hepatic impairment (Child-Pugh B), and 20-30% higher in moderate-to-severe renal impairment (CLcr <50 mL/min); severe hepatic impairment (Child-Pugh C) was never studied. Strong CYP3A4 inhibitors dramatically prolong the half-life — ritonavir extended it to 26 hours and indinavir roughly doubled it — which is why co-dosing carries hard dose caps.
